Overview

Simeprevir (TMC435) is an oral small molecule inhibitor of the hepatitis C virus (HCV) NS3/4A protease, developed for the treatment of chronic HCV genotype 1 infection. It is marketed under the brand name Olysio and was developed by Janssen in collaboration with Medivir[6][8]. Rilpivirine (TMC278) is a diarylpyrimidine nonnucleoside reverse transcriptase inhibitor (NNRTI), used as an antiretroviral agent for HIV-1 infection. It acts by inhibiting HIV-1 reverse transcriptase and has a high genetic barrier to resistance[4][5][7]. Rilpivirine is marketed as Edurant and was also developed by Janssen. The combination of these two drugs represents a multi-antiviral regimen targeting both HCV and HIV infections.
