Overview

siMLK4 is a small interfering RNA (siRNA) designed to target and knock down the expression of Mixed Lineage Kinase 4 (MLK4, also known as MAP3K21), a member of the MAP3K family. In preclinical research, siMLK4 has been utilized to investigate the role of MLK4 in cancer biology, particularly in triple-negative breast cancer (TNBC) and HER2-positive, Herceptin-resistant breast cancers. Knockdown of MLK4 using siMLK4 or siMLK4-loaded nanoparticles has been shown to induce cell cycle arrest, promote apoptosis, and sensitize resistant breast cancer cells to therapies such as Herceptin (trastuzumab) and chemotherapeutic agents like doxorubicin. It is primarily used as a research reagent to validate MLK4 as a therapeutic target.
