**Simnotrelvir + ritonavir** is an oral, co-packaged small-molecule antiviral product for treatment of mild to moderate COVID-19 in adults. Simnotrelvir is a peptidomimetic inhibitor of the SARS-CoV-2 main protease, also called 3C-like protease, which blocks viral polyprotein processing and thereby prevents viral replication. Ritonavir is included as a pharmacokinetic enhancer that inhibits CYP3A-mediated simnotrelvir metabolism, increasing simnotrelvir exposure. The product is supplied as separate simnotrelvir and ritonavir tablets for concomitant oral administration; the labeled standard regimen is simnotrelvir 750 mg plus ritonavir 100 mg every 12 hours for 5 days. It was jointly developed by the Shanghai Institute of Materia Medica, Wuhan Institute of Virology, and Simcere, and received regular NMPA approval in China for adult mild to moderate COVID-19 in July 2024.
