Overview

Simnotrelvir (SIM0417, also known as SSD8432) is an oral small molecule inhibitor of the SARS-CoV-2 main protease (3CLpro), developed for the treatment of COVID-19. It acts by inhibiting viral replication through blockade of the 3-chymotrypsin-like protease essential for coronavirus maturation and propagation. Ritonavir is co-administered as a pharmacokinetic booster to inhibit CYP3A4-mediated metabolism and increase systemic exposure to simnotrelvir. Itraconazole is a triazole antifungal agent that strongly inhibits CYP3A4 and may further affect the pharmacokinetics of both simnotrelvir and ritonavir when used in combination. This triple combination has been studied in drug-drug interaction trials to evaluate safety and pharmacokinetic effects in healthy volunteers[1][2][5].
