Simplexia's HSV-2 vaccine is a protein subunit vaccine candidate currently in preclinical development for the prevention and treatment of genital herpes (HSV-2 infection). The vaccine specifically targets the HSV-2-specific glycoprotein G-2 (gG-2), utilizing a truncated, soluble version of the mature membrane-bound region known as mgG-2 (specifically the EXCT4 antigen form). Its unique mechanism of action involves inducing antibodies that block gG-2, which is essential for the virus's ability to spread from cell to cell. By inhibiting this protein, the vaccine "locks down" the virus at the cell membrane, preventing transmission and spread. Developed based on research from the University of Gothenburg, the vaccine has shown the ability to elicit Th1-polarized CD4+ T cell responses and antibodies with antibody-dependent cellular cytotoxicity (ADCC) in preclinical models.
