Simplirix (GSK208141) is a protein-based viral subunit vaccine developed for the prevention of genital herpes caused by herpes simplex virus type 2 (HSV-2). The active component is a truncated form of the HSV-2 glycoprotein D (gD), formulated with an adjuvant to enhance immunogenicity. The mechanism of action involves immunostimulation—eliciting an immune response against HSV-2 to prevent infection. Developed by GlaxoSmithKline in collaboration with the National Institute of Allergy and Infectious Diseases and originally originated at Genentech. Despite reaching phase III clinical trials for prevention of genital herpes in seronegative individuals and those at risk for HSV infection in multiple regions including the US and EU, development was discontinued after failing to demonstrate sufficient efficacy[1][4].
