Overview
**Simplirix** is an investigational prophylactic herpes simplex vaccine developed by GSK to prevent genital herpes. It is a protein-subunit vaccine containing truncated herpes simplex virus type 2 glycoprotein D antigen formulated with the AS04 adjuvant system, comprising aluminum hydroxide and 3-O-deacylated monophosphoryl lipid A. The vaccine was designed to induce glycoprotein D-specific humoral and cellular immunity that could prevent herpes simplex virus entry and genital disease. In the Phase 3 HerpeVac Trial for Women, the primary endpoint for prevention of genital herpes was not met, and GSK discontinued further development.
