Overview

Simridarlimab is a bispecific humanized monoclonal antibody that simultaneously targets the innate immune checkpoint CD47 and the adaptive immune checkpoint programmed death-ligand 1 (PD-L1/CD274), designed to enhance antitumor immunity by promoting macrophage-mediated phagocytosis while blocking PD-1/PD-L1 signaling in the tumor microenvironment.[1][3][5][10][11] The molecule is engineered as a mixed mAb/VH-VH-CH2-CH3 bispecific format, with an anti-CD47 IgG1 arm and a single-domain anti–PD-L1 arm, and shows preferential binding to CD47+PD-L1+ tumor cells to limit red blood cell binding and on-target hematologic toxicity.[1][3][10][11] Developed by Innovent Biologics, simridarlimab is being investigated primarily in oncology, including advanced solid tumors, hematologic malignancies, lymphoma, myeloid tumors, classical Hodgkin lymphoma resistant to prior PD-1/PD-L1 blockade, and non-small cell lung cancer (including ALK-rearranged disease), where early-phase trials have shown manageable safety and preliminary antitumor activity.[2][3][7][10][15]
