Simvastatin + losartan + amlodipine + hydrochlorothiazide is a four-component cardiovascular polypill developed for the primary prevention of cardiovascular disease (CVD) in individuals aged 50 and older without a prior history of CVD. Developed by researchers at the Wolfson Institute of Preventive Medicine (Queen Mary University of London), the combination includes simvastatin (40 mg), losartan (25 mg), amlodipine (2.5 mg), and hydrochlorothiazide (12.5 mg) in a single daily tablet. The pill aims to reduce the risk of heart attacks and strokes by simultaneously lowering LDL cholesterol and blood pressure. It was evaluated in the Phase III Polypill Prevention Trial 1, a randomized, double-blind, placebo-controlled crossover trial, which demonstrated significant reductions in systolic and diastolic blood pressure as well as LDL cholesterol levels.
