This is a combination of three small-molecule drugs—simvastatin, spironolactone, and amlodipine. Simvastatin is an HMG-CoA reductase inhibitor (statin) used to lower cholesterol and reduce cardiovascular risk. Spironolactone is a potassium-sparing diuretic and aldosterone antagonist used for hypertension, heart failure, edema, and hyperaldosteronism. Amlodipine is a calcium channel blocker indicated for hypertension and angina. The combination would theoretically target both lipid lowering (simvastatin) and blood pressure control through two mechanisms (spironolactone’s diuretic/aldosterone antagonism; amlodipine’s vasodilation via L-type calcium channel inhibition). There are no known fixed-dose or branded products containing all three agents together; this combination may be prescribed off-label in select patients with comorbid hypertension, heart failure or edema, and dyslipidemia who require all three mechanisms of action[1][5][7]. Notably, there are clinically significant drug-drug interactions between simvastatin and amlodipine that require dose adjustment due to increased risk of statin toxicity[6][7][8].
