Overview

Sing2016 M2SR H3N2 is an investigational intranasal influenza vaccine based on a live but replication-deficient virus platform called "M2-deficient single replication" (M2SR). The vaccine is engineered by deleting the gene for the viral matrix protein 2 (M2), which is essential for viral replication. During manufacturing, the missing protein is supplied in trans to allow production of the vaccine virus; however, after administration to humans it can only undergo a single round of replication and cannot produce infectious progeny[8][10]. This design allows robust stimulation of both mucosal and systemic immune responses—including antibodies against hemagglutinin (HA), neuraminidase (NA), nucleoprotein (NP), and other conserved antigens—while minimizing safety risks associated with replicating viruses[7][8][10]. The "Sing2016" designation refers to its use of HA and NA genes from the A/Singapore/INFIMH-16-0019/2016 strain of influenza A/H3N2[10]. Developed primarily as a prophylactic against seasonal influenza infection caused by drifted or matched strains of H3N2, it has been tested in adults and children for safety and immunogenicity[4][5][9].
