Overview

AmideBio's single-chain insulin (SCI), also known as Ultrastable Insulin, is a peptide-based insulin analogue designed for enhanced chemical and thermal stability. Unlike native insulin, which consists of two chains (A and B) held together by disulfide bonds, SCI is a single polypeptide chain where the B and A chains are connected by a short joining peptide (typically 5 to 9 amino acids). This structural modification renders the molecule resistant to fibrillation and deamidation, allowing it to remain stable at 25°C for up to three months. It acts as an insulin receptor agonist with potency and selectivity similar to native human insulin. The drug is currently in preclinical development for the treatment of diabetes mellitus, specifically targeting use in insulin pumps and regions with limited refrigeration infrastructure.
