Drug intelligence / Profile preview
Single-domain antibody (sdAb) BCMA CAR-T cells are chimeric antigen receptor T-cell therapies engineered to target B-cell maturation antigen (BCMA), a protein highly expressed on malignant plasma cells in multiple myeloma. Unlike conventional CAR-T cells that utilize single-chain variable fragments (scFvs) as their antigen-binding domains, these constructs employ camelid-derived single-domain antibodies (also known as nanobodies or VHHs). Due to their smaller size, high stability, and lower propensity for tonic signaling, sdAb-based CARs can potentially offer enhanced tumor-killing efficacy, improved persistence, and reduced T-cell exhaustion. Preclinical development of these Canadian-made constructs, such as sdBCMA.41BB.CD3z and sdBCMA1.41BB.CD3z, is being led by the BC Cancer Research Institute, the National Research Council of Canada, and the Mayo Clinic, with evaluations ongoing in immunocompetent and xenograft models of multiple myeloma.
