Drug intelligence / Profile preview
Sintilimab + rituximab + lenalidomide is a chemotherapy-free triplet regimen combining the anti‑PD‑1 monoclonal antibody sintilimab, the anti‑CD20 monoclonal antibody rituximab, and the immunomodulatory small molecule lenalidomide, under evaluation primarily for diffuse large B‑cell lymphoma (DLBCL) in older or unfit patients. Sintilimab blocks the programmed cell death 1 (PD‑1) receptor on T cells to restore antitumor immunity, rituximab targets CD20 on B cells to induce antibody‑dependent cellular cytotoxicity and complement‑mediated lysis, and lenalidomide enhances T‑ and NK‑cell function, modulates the tumor microenvironment, and exerts direct antiproliferative activity on malignant B cells. Early phase 2 data with this sR2 combination suggest meaningful activity and tolerability as a first‑line, chemotherapy‑free option in de novo DLBCL, with ongoing development focused on optimizing immunotherapy‑based regimens for B‑cell lymphomas.[4][5]
