This is a multi-agent combination regimen consisting of sintilimab, anlotinib, pemetrexed, and either cisplatin or carboplatin. Sintilimab is a monoclonal antibody targeting programmed cell death protein 1 (PD-1), functioning as an immune checkpoint inhibitor to enhance anti-tumor immune responses. Anlotinib is a small molecule tyrosine kinase inhibitor with antiangiogenic properties, targeting VEGFR 1/2/3, FGFR1–4, PDGFR α/β, and c-Kit. This four-drug regimen has been investigated as first-line therapy in advanced or metastatic pleural mesothelioma and non-small cell lung cancer (NSCLC), aiming to combine immunotherapy with antiangiogenic therapy and cytotoxic chemotherapy for synergistic antitumor effects[3][4]. The regimen leverages distinct mechanisms—immune modulation by sintilimab, angiogenesis inhibition by anlotinib, antimetabolite action by pemetrexed, and DNA damage induction by platinum agents—to maximize efficacy against aggressive thoracic malignancies.
