Sintilimab + arsenic trioxide is an investigational combination therapy being evaluated for the treatment of advanced intrahepatic cholangiocarcinoma (ICC). Sintilimab is a recombinant humanized IgG4 monoclonal antibody that binds to the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2, thereby restoring T-cell antitumor activity. Arsenic trioxide (ATO) is a small molecule that induces apoptosis in cancer cells and has been shown to modulate the tumor microenvironment, potentially enhancing the efficacy of immune checkpoint inhibitors by inhibiting angiogenesis and inducing reactive oxygen species. This combination is being pioneered by researchers at the Third Affiliated Hospital of Naval Medical University (Eastern Hepatobiliary Surgery Hospital) to address the poor prognosis and limited treatment options for ICC.
