Overview

This combination therapy is an investigational first-line regimen for locally advanced or metastatic cholangiocarcinoma, integrating immunotherapy, anti-angiogenic therapy, and cytotoxic chemotherapy. Sintilimab is a humanized monoclonal antibody that blocks the interaction between PD-1 and its ligands (PD-L1 and PD-L2), thereby restoring T-cell antitumor activity. Bevacizumab is a monoclonal antibody that targets VEGF-A, inhibiting angiogenesis and potentially normalizing tumor vasculature to improve drug delivery and immune cell infiltration. The AG regimen consists of albumin-bound paclitaxel (nab-paclitaxel), a microtubule stabilizer, and gemcitabine, a nucleoside analog that inhibits DNA synthesis. This multi-pronged approach aims to enhance the objective response rate and survival outcomes in patients with biliary tract cancers.
