This combination therapy is an antineoplastic regimen primarily used for the treatment of advanced non-squamous non-small cell lung cancer (nsNSCLC). It combines **sintilimab**, a recombinant humanized IgG4 monoclonal antibody that binds to the programmed cell death protein 1 (PD-1) receptor to restore T-cell activity, with **bevacizumab**, a monoclonal antibody that targets vascular endothelial growth factor (VEGF) to inhibit angiogenesis. The addition of platinum-based **chemotherapy** (typically pemetrexed and cisplatin or carboplatin) provides direct cytotoxic effects and promotes the release of tumor antigens. This quadruple regimen has shown significant efficacy, particularly in patients with EGFR-mutated nsNSCLC who have progressed after tyrosine kinase inhibitor (TKI) therapy, as demonstrated in the ORIENT-31 phase 3 trial. The combination is designed to leverage the synergistic effects of immune checkpoint inhibition and anti-angiogenesis to overcome resistance in the tumor microenvironment.
