This is a combination regimen consisting of the anti-PD-1 monoclonal antibody sintilimab with either carboplatin or cisplatin (both platinum-based chemotherapies) and the antifolate agent pemetrexed. Sintilimab is an immune checkpoint inhibitor that blocks programmed cell death protein 1 (PD-1), thereby enhancing T-cell mediated antitumor responses. Carboplatin and cisplatin are DNA-damaging agents that induce apoptosis in rapidly dividing cells, while pemetrexed inhibits folate-dependent enzymes involved in nucleotide synthesis, disrupting DNA replication in cancer cells. This combination has been investigated as a first-line treatment for advanced or metastatic nonsquamous non-small cell lung cancer (NSCLC) without EGFR mutations or ALK rearrangements[2][3][4][5].
