Sintilimab + gemcitabine + paclitaxel is a combination regimen used in oncology clinical trials for the treatment of various advanced cancers. Sintilimab is a fully humanized IgG4 monoclonal antibody that targets Programmed cell death protein 1 (PD-1), blocking its interaction with PD-L1 and PD-L2 to relieve immunosuppression and enhance T-cell mediated anti-tumor activity[6]. Gemcitabine is a nucleoside analog that inhibits DNA synthesis by incorporating into DNA strands during replication. Paclitaxel (including its albumin-bound form nab-paclitaxel) stabilizes microtubules and prevents their depolymerization during cell division, leading to apoptosis of cancer cells. The combination leverages the synergistic effects of chemotherapy-induced immunogenic cell death with immune checkpoint blockade to improve anti-tumor efficacy[1][2][3][4][5][6]. This regimen has been investigated in phase II studies for diseases such as metastatic pancreatic adenocarcinoma and unresectable locally advanced pancreatic cancer[2][8], as well as other solid tumors.
