Overview

Sintilimab (IBI308) is a recombinant human IgG4 monoclonal antibody that binds to the PD-1 receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell mediated anti-tumor activity. In this combination therapy, it is paired with irinotecan, a topoisomerase I inhibitor, and 5-fluorouracil (5-FU), an antimetabolite that inhibits thymidylate synthase. This chemo-immunotherapy regimen is being developed by Innovent Biologics, in collaboration with Eli Lilly, for the treatment of various advanced malignancies, including non-small cell lung cancer and gastrointestinal cancers. The combination aims to leverage the cytotoxic effects of chemotherapy to enhance the immune-mediated response of the checkpoint inhibitor.
