Sintilimab + mFFN + NALIRIFOX is a combination chemo-immunotherapy regimen being investigated for the treatment of unresectable locally advanced pancreatic cancer. The regimen consists of sintilimab, a recombinant humanized IgG4 monoclonal antibody that binds to the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell mediated anti-tumor activity. This is combined with one of two chemotherapy backbones: modified FOLFIRINOX (mFFN), which includes fluorouracil, leucovorin, irinotecan, and oxaliplatin; or NALIRIFOX, which substitutes standard irinotecan with nanoliposomal irinotecan. The regimen is typically administered in conjunction with radiotherapy. It is currently being evaluated in a Phase II clinical trial sponsored by the Shandong Cancer Hospital and Institute.
