This combination regimen is a neoadjuvant therapy being investigated for early-stage, treatment-naive triple-negative breast cancer (TNBC). It consists of the PD-1 inhibitor sintilimab administered alongside a sequential chemotherapy backbone. The chemotherapy includes four cycles of albumin-bound paclitaxel (nab-paclitaxel) and carboplatin, followed by four cycles of epirubicin and cyclophosphamide. Sintilimab is a recombinant humanized IgG4 monoclonal antibody that binds to the PD-1 receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell antitumor activity. This regimen is often studied in combination with stereotactic body radiotherapy (SBRT) to enhance immunogenicity and improve pathological complete response (pCR) rates in high-risk TNBC patients.
