Overview

This combination therapy is a chemo-immunotherapy regimen comprising the anti-PD-1 monoclonal antibody sintilimab, the albumin-bound microtubule inhibitor nab-paclitaxel, and a platinum-based chemotherapeutic agent (typically cisplatin or carboplatin). Sintilimab, developed by Innovent Biologics and Eli Lilly, functions as a checkpoint inhibitor by blocking the interaction between the Programmed Cell Death Protein 1 (PD-1) receptor on T cells and its ligands (PD-L1 and PD-L2), thereby reactivating the immune system's anti-tumor response. Nab-paclitaxel and platinum agents provide direct cytotoxic effects; nab-paclitaxel inhibits microtubule dynamics to prevent cell division, while platinum agents induce DNA cross-linking and apoptosis. This regimen is currently being investigated in a multicenter, randomized Phase III clinical trial (led by Qunxing Li, MD) as a perioperative (neoadjuvant and adjuvant) treatment strategy for patients with locally advanced oral squamous cell carcinoma (OSCC) to improve event-free survival and pathological response rates.
