This is a combination therapy consisting of sintilimab (an anti-PD-1 monoclonal antibody), nab-paclitaxel (a novel taxane formulation), and S-1 (a combination of tegafur, gimeracil, and oteracil). Sintilimab blocks the interactions between PD-1 and its ligands, enhancing T-cell immune responses against tumor cells. Nab-paclitaxel is an albumin-bound formulation of paclitaxel with advantages including less hypersensitivity reactions, neurotoxicity, and neutropenia compared to conventional paclitaxel. S-1 is an oral fluoropyrimidine that combines tegafur (a prodrug of 5-fluorouracil), gimeracil (which inhibits 5-FU degradation), and oteracil (which reduces gastrointestinal toxicity). This triplet regimen is being investigated for various cancers including gastric cancer and biliary tract carcinoma.
