Overview

This combination therapy consists of the PD-1 inhibitor sintilimab and the quadrivalent human papillomavirus (HPV) vaccine (Gardasil). It is being investigated as a novel therapeutic approach for patients with recurrent or metastatic cervical cancer who have failed or are intolerant to standard first-line treatments. Sintilimab is a monoclonal antibody that blocks the PD-1 receptor, thereby restoring T-cell mediated anti-tumor immunity. Gardasil is a recombinant vaccine containing virus-like particles (VLPs) of the L1 capsid protein from HPV types 6, 11, 16, and 18. In this therapeutic setting, the vaccine is intended to stimulate an HPV-specific immune response that is further amplified by the checkpoint inhibition provided by sintilimab. A Phase II clinical trial led by Buhai Wang demonstrated promising results, including a 53.8% objective response rate and a 76.9% disease control rate in patients with advanced cervical cancer.
