Overview

The regimen integrates the PD-1 checkpoint inhibitor sintilimab, the multi-kinase inhibitor regorafenib, and hepatic arterial infusion chemotherapy (HAIC) utilizing the FOLFOX protocol (oxaliplatin, fluorouracil, and leucovorin). Sintilimab works by restoring T-cell mediated anti-tumor activity, while regorafenib inhibits multiple kinases involved in angiogenesis and oncogenesis. The HAIC component delivers high concentrations of cytotoxic agents directly to the liver via the hepatic artery, aiming to maximize local tumor control and overcome systemic treatment resistance in patients who have failed standard second-line therapies.
