Overview

Sintilimab + sorafenib is a combination therapy consisting of a programmed cell death protein 1 (PD-1) inhibitor and a multi-kinase inhibitor. Sintilimab, developed by Innovent Biologics and Eli Lilly, is a humanized immunoglobulin G4 (IgG4) monoclonal antibody that binds to the PD-1 receptor on T cells, blocking its interaction with PD-L1 and PD-L2 to restore anti-tumor immune activity. Sorafenib, developed by Bayer and Onyx Pharmaceuticals, is an oral small-molecule inhibitor that targets multiple intracellular and cell surface kinases, including Raf kinases (C-Raf and B-Raf), VEGFR-1, VEGFR-2, VEGFR-3, and PDGFR-beta. By inhibiting these pathways, sorafenib suppresses tumor cell proliferation and angiogenesis. This combination is primarily investigated for the treatment of advanced hepatocellular carcinoma (HCC), particularly in clinical trials conducted by institutions such as the Cancer Hospital of the Chinese Academy of Medical Sciences, often exploring its efficacy as a systemic therapy or in combination with interventional procedures like transarterial chemoembolization (TACE).
