Overview

sintilimab + SOX is a combination therapy used primarily in the treatment of advanced or locally advanced gastric cancer. It consists of sintilimab, a programmed cell death protein 1 (PD-1) immune checkpoint inhibitor (monoclonal antibody), and the SOX chemotherapy regimen. The SOX regimen includes S-1 (an oral fluoropyrimidine: a combination of tegafur, gimeracil, and oteracil) and oxaliplatin (a platinum-based chemotherapeutic). This combination is being evaluated in perioperative and first-line settings for gastric cancer, showing higher pathological response rates compared to chemotherapy alone in several clinical trials. The mechanism of action relies on both direct cytotoxic effects (via oxaliplatin and S-1) and immune-mediated anti-tumor activity (via sintilimab targeting PD-1, restoring T-cell mediated cancer cell killing)[1][3][4][9].
