sintilimab + taxane + carboplatin

A combination neoadjuvant regimen consisting of the PD-1 inhibitor sintilimab and a chemotherapy backbone of a taxane (such as nab-paclitaxel, docetaxel, or paclitaxel) and carboplatin. This specific therapeutic approach is being evaluated in the NeoSTEP trial (NCT05843292), sponsored by the Shanghai Jiao Tong University School of Medicine, for the treatment of early-stage triple-negative breast cancer (TNBC). The regimen involves a short-term administration of sintilimab (2 cycles) in conjunction with 4 cycles of taxane and carboplatin chemotherapy prior to surgery. The study aims to assess whether this abbreviated immunotherapy schedule can maintain high pathological complete response (pCR) rates while reducing the incidence of immune-related adverse events typically associated with longer-term checkpoint inhibition.
