SINV-GM-CSF is a recombinant oncolytic Sindbis virus (SINV) engineered to express human granulocyte-macrophage colony-stimulating factor (GM-CSF). The viral vector incorporates a specific G285S mutation in the nsp1 protein, which is designed to enhance the therapeutic profile and safety of the vector for cancer treatment. As an oncolytic virus, SINV-GM-CSF selectively infects and induces immunogenic cell death in tumor cells, such as hepatocellular carcinoma (HCC). The inclusion of the GM-CSF transgene serves to modulate the tumor microenvironment by recruiting and activating immune cells, specifically promoting the polarization of anti-tumor M1-type macrophages while reducing pro-tumor M2-type macrophages. This dual mechanism of direct viral oncolysis and localized cytokine-mediated immune stimulation aims to overcome the immunosuppressive environment of refractory tumors.
