Overview

SION-451 is a novel small molecule drug developed by Sionna Therapeutics for the treatment of cystic fibrosis. It acts as a stabilizer of the nucleotide-binding domain 1 (NBD1) of the cystic fibrosis transmembrane conductance regulator (CFTR) protein, aiming to restore or enhance CFTR function in patients with cystic fibrosis, particularly those with mutations such as F508del. By stabilizing NBD1, SION-451 helps correct misfolding and dysfunction of CFTR, potentially improving chloride transport and clinical outcomes. The drug has demonstrated high-affinity binding to NBD1 and increased stability of defective CFTR protein in preclinical models. Phase 1 clinical trials have shown that SION-451 is generally well tolerated in healthy volunteers, achieving desired pharmacokinetic targets without serious adverse events. Future development includes combination studies with other proprietary modulators targeting different domains of CFTR[2][3][4][5][6][7][8][9].
