KW-6356 (sipagladenant) is a selective, non-xanthine antagonist and inverse agonist of the adenosine A2A receptor. Developed by Kyowa Kirin for the treatment of Parkinson's disease, it was designed as a second-generation A2A receptor antagonist with higher affinity and selectivity compared to earlier agents. The drug demonstrated efficacy in improving motor symptoms both as monotherapy and as adjunctive therapy with levodopa in Phase 2 clinical trials for Parkinson’s disease. Despite promising results regarding safety and efficacy in these studies, development was discontinued by Kyowa Kirin[1][3][4][5][6].
