Drug intelligence / Profile preview
siPlasminogen (also known as SG-001 or siPLG) is a preclinical-stage, long-acting, liver-targeted small interfering RNA (siRNA) therapeutic designed to inhibit fibrinolysis and reduce bleeding in patients with bleeding disorders, such as hemophilia A and von Willebrand disease. Developed by SeraGene Therapeutics, a spin-out from the Versiti Blood Research Institute and the University of British Columbia, siPlasminogen is encapsulated in lipid nanoparticles (LNPs) for intravenous administration. It works by precisely knocking down the expression of plasminogen, the central protein of the fibrinolytic pathway, thereby stabilizing blood clots and restoring the balance between coagulation and fibrinolysis. In preclinical models, including canine models of hemophilia A, siPlasminogen has demonstrated the potential to significantly reduce bleeding events and the need for factor infusions with a prolonged duration of action, offering a promising long-term prophylactic alternative to short-acting antifibrinolytics like tranexamic acid.
