Overview

Sipoglitazar (TAK-654) is a novel, orally active pan-peroxisome proliferator-activated receptor (PPAR) agonist that targets the alpha (α), delta (δ), and gamma (γ) isoforms. Developed by Takeda Pharmaceuticals, it was primarily investigated for the treatment of type 2 diabetes mellitus and associated metabolic disorders. By activating the PPAR family of nuclear receptors, sipoglitazar regulates the expression of genes involved in glucose and lipid metabolism, aiming to improve insulin sensitivity and lipid profiles. Although it reached clinical development, Takeda eventually discontinued the program.
