Overview

Sipucibizumab (also referred to in some clinical trial contexts as spicichibant) is a humanized monoclonal antibody that targets the interleukin-4 receptor alpha (IL-4Rα) subunit. By binding to IL-4Rα, the drug inhibits the signaling of both interleukin-4 (IL-4) and interleukin-13 (IL-13), which are critical cytokines in the Type 2 inflammatory pathway. This mechanism is designed to suppress the allergic inflammatory cascade associated with conditions such as allergic rhinitis and atopic dermatitis. Sipucibizumab is being evaluated as an intranasal formulation (nasal spray) for the treatment of moderate to severe perennial allergic rhinitis, with clinical studies focusing on its ability to improve nasal mucociliary transit time, symptom-medication scores, and overall quality of life.
