SIR1-365 is a potent and selective small molecule inhibitor of receptor-interacting serine/threonine-protein kinase 1 (RIPK1). RIPK1 is a key mediator in cell death pathways, including apoptosis and necroptosis, as well as inflammation triggered by pro-inflammatory cytokines and pathogens such as SARS-CoV-2. By inhibiting RIPK1, SIR1-365 aims to reduce excessive inflammation and inflammatory cell death associated with severe diseases like COVID-19. The drug has been evaluated primarily for its safety and efficacy in hospitalized patients with severe COVID-19, where it demonstrated good tolerability, reduced incidence of respiratory failure, improved arterial oxygenation, shortened hospitalization duration, and showed trends toward faster clinical recovery compared to placebo. It has also been investigated for systemic inflammatory response syndrome (SIRS), neurodegenerative diseases (including Parkinson’s disease), nerve degeneration, kidney diseases, multiple sclerosis, Alzheimer’s disease, and general inflammation[2][3][5][7][8].
