Sirexatamab + pembrolizumab is an investigational combination therapy being developed by Leap Therapeutics for the treatment of advanced or recurrent endometrial cancer. Sirexatamab (DKN-01) is a humanized monoclonal antibody that targets Dickkopf-related protein 1 (DKK1), a secreted protein that modulates the Wnt signaling pathway and is associated with tumor growth and immune suppression. Pembrolizumab is a well-established anti-PD-1 checkpoint inhibitor. The combination aims to synergistically enhance anti-tumor immune responses by simultaneously inhibiting DKK1-mediated immune evasion and restoring T-cell activity through PD-1 blockade. This regimen is currently being evaluated in a Phase 2 clinical trial (NCT05761951) specifically in patients with DKK1-high endometrial tumors.
