Sirf CAR-T cells

**Sirf CAR-T cells** are an academic, preclinical armored CAR-T-cell product designed for solid tumors. Murine T cells were engineered with an anti-Trop2 CAR and to secrete a signal regulatory protein alpha Fc fusion protein. The CAR mediates Trop2-directed tumor-cell recognition and cytotoxicity, while locally secreted SIRPα-Fc binds CD47 and disrupts the CD47-SIRPα macrophage checkpoint, enhancing phagocytosis of tumor cells. In syngeneic immunocompetent mouse models of colon and breast cancer, the construct reduced tumor burden, prolonged survival, improved intratumoral CAR-T persistence, increased central-memory T cells and pro-inflammatory macrophage features, and reduced immunosuppressive myeloid cells.
