Overview

siRNA (CLPTM1L) is a therapeutic candidate utilizing small interfering RNA (siRNA) technology to target the Cleft Lip and Palate Transmembrane 1-Like protein (CLPTM1L). CLPTM1L is an endoplasmic reticulum (ER) stress-inducible oncofetal protein that is overexpressed on the plasma membrane of multiple myeloma cells and associated with chemoresistance. By silencing CLPTM1L expression, this siRNA inhibits the AKT signaling pathway, leading to the downregulation of anti-apoptotic proteins (BCL2, BCLXL) and the upregulation of pro-apoptotic markers (cleaved caspase-3, BID, cleaved PARP). Preclinical studies indicate that CLPTM1L knockdown sensitizes myeloma cells to proteasome inhibitors such as bortezomib, suggesting its potential as a combination therapy for resistant blood cancers.
