siRNA against LOXL3 is an experimental RNA interference (RNAi) agent designed to specifically target and silence the expression of the Lysyl Oxidase-Like 3 (LOXL3) gene. LOXL3 is a member of the lysyl oxidase family of enzymes, which are responsible for catalyzing the cross-linking of collagen and elastin in the extracellular matrix. In the context of neuro-oncology, LOXL3 has been identified as being significantly upregulated in high-grade astrocytomas, including glioblastoma (GBM), where its expression correlates with poor patient prognosis. Preclinical research using siRNA-mediated knockdown of LOXL3 in mesenchymal GBM cell lines (such as U87MG) has demonstrated a reduction in cell proliferation and an increase in apoptosis, particularly when used in combination with the chemotherapy agent temozolomide. Furthermore, LOXL3 silencing appears to disrupt mitochondrial dynamics, leading to morphological changes in mitochondria and a decrease in mitochondrial DNA copy number, suggesting a role for LOXL3 in maintaining mitochondrial stability in cancer cells.
