Overview

siRNA against RRM2 refers to small interfering RNA (siRNA) molecules designed to target and silence the expression of the ribonucleotide reductase subunit M2 (RRM2) gene. RRM2 is a critical subunit of ribonucleotide reductase, an enzyme that catalyzes the conversion of ribonucleotides to deoxyribonucleotides, which is a rate-limiting step essential for DNA synthesis and replication. Because RRM2 is frequently overexpressed in various cancers and is associated with tumor progression, invasion, and resistance to chemotherapies (such as gemcitabine and doxorubicin), silencing RRM2 via RNA interference (RNAi) represents a promising therapeutic strategy. Various academic groups and companies have investigated RRM2-targeting siRNAs, often formulated in lipid nanoparticles (LNPs) or other nanoparticle delivery systems (such as the transferrin-targeted cyclodextrin nanoparticle CALAA-01), for the treatment of solid tumors including pancreatic cancer, hepatocellular carcinoma, melanoma, and head and neck cancers.
