Overview

siRNA-ATM is a small interfering RNA (siRNA) designed to silence the expression of the ataxia-telangiectasia mutated (ATM) gene. ATM is a serine/threonine protein kinase that acts as a master regulator of the DNA damage response (DDR), particularly in response to DNA double-strand breaks caused by ionizing radiation. By utilizing the RNA interference (RNAi) pathway to degrade ATM mRNA, siRNA-ATM reduces ATM protein levels, thereby impairing the cell's ability to repair DNA damage. This mechanism is primarily explored as a strategy to overcome radioresistance in cancers such as glioblastoma, where glioma stem cells (GSCs) often exhibit high ATM expression. Preclinical research indicates that silencing ATM increases radiation-induced apoptosis and G2 phase arrest, effectively sensitizing tumor cells to radiotherapy.
