siRNA B7-H1-2 is an experimental small interfering RNA (siRNA) sequence designed to target and silence the expression of B7-H1 (also known as Programmed death-ligand 1, PD-L1, or CD274). It functions through the RNA interference (RNAi) pathway, where it binds to and triggers the degradation of B7-H1 messenger RNA (mRNA), thereby preventing the translation of the B7-H1 protein. In preclinical studies involving colorectal cancer cell lines such as HCT116, siRNA B7-H1-2 has been utilized to demonstrate that B7-H1 knockdown leads to significant inhibition of cell proliferation, migration, and invasion, as well as an increase in apoptosis. These findings suggest that B7-H1 may play a direct role in promoting tumor cell aggressiveness and survival, making it a potential therapeutic target beyond its established role in immune evasion.
