siRNA CD147 is a small interfering RNA (siRNA) therapeutic candidate designed to silence the expression of CD147, also known as Basigin or extracellular matrix metalloproteinase inducer (EMMPRIN). CD147 is a transmembrane glycoprotein that is highly expressed in various cancers, including glioblastoma multiforme (GBM), where its expression levels correlate with reduced overall survival. In glioma research, CD147 has been identified as a downstream effector of Galectin-1 (Gal-1), regulating the expression of matrix metalloproteinase-9 (MMP9) and integrins, which are critical for tumor progression and the maintenance of glioma stem cells. By specifically degrading CD147 mRNA, this siRNA inhibits the Gal-1-CD147-MMP9 signaling pathway, potentially reducing tumor invasiveness and progression. Experimental studies in glioma stem cell lines and mouse brain models have demonstrated that siRNA-mediated knockdown of CD147 significantly reduces protein levels and downstream MMP9 activity.
