siRNA MLF2 is an experimental, research-stage small interfering RNA (siRNA) therapeutic designed to target and silence the myeloid leukemia factor 2 (MLF2) gene. Developed primarily by researchers at the Houston Methodist Research Institute and Weill Cornell Medicine, siRNA MLF2 works by inhibiting MLF2 expression, which has been identified as a key driver of tumor initiation, metastasis, and nitric oxide (NO) signaling pathways in triple-negative breast cancer (TNBC). In preclinical studies, siRNA MLF2, often delivered via a multistage vector (MSV) nanoparticle platform and evaluated in combination with siRNA RPL39 or chemotherapy, significantly reduced tumor volume, lung metastases, and breast cancer stem cell (BCSC) markers in patient-derived xenograft (PDX) models.
