**siRNA-PD-1** refers to a chemically synthesized small interfering RNA (siRNA) or self-delivering RNA (sdRNA) molecule designed to specifically target and knock down the mRNA of the *programmed cell death protein 1* (PD-1, encoded by the PDCD1 gene). By silencing PD-1 expression, siRNA-PD-1 blocks this immune checkpoint, which is otherwise exploited by tumors to evade immune detection. siRNA-PD-1 has been used in multiple experimental delivery approaches, including encapsulation within solid lipid nanoparticles (SLNs), chemical backbone modification for self-delivery (sdRNA), and other vehicles, for both direct immunomodulation (in macrophages or T cells) and ex vivo cell therapy (TIL expansion). Its principal mechanism is to restore or boost immune cell activity against tumors by preventing expression of the PD-1 checkpoint, and it has demonstrated antitumor activity in mouse models and ex vivo in human T cells[1][2].
