Overview

siRNA RPL39 is an investigational small interfering RNA (siRNA) therapeutic designed to target and silence the expression of the RPL39 gene, which encodes Ribosomal Protein L39 (a component of the 60S ribosomal subunit). Developed primarily by researchers at Houston Methodist and Weill Cornell Medicine, siRNA RPL39 is being evaluated in preclinical studies for the treatment of triple-negative breast cancer (TNBC), metaplastic breast cancer (MpBC), and pancreatic cancer. By silencing RPL39, the drug suppresses tumor cell proliferation, induces apoptosis, and reduces metastasis, particularly by inhibiting nitric oxide synthase (NOS) signaling and reducing breast cancer stem cell self-renewal. Various nanoparticle-based delivery systems, including multistage vector (MSV) liposomal nanoparticles and biomimetic leukosomes, have been utilized to facilitate its in vivo delivery.
