Drug intelligence / Profile preview
siRNA targeting Apex1 is a small interfering RNA (siRNA) therapeutic designed to silence the expression of apurinic/apyrimidinic endodeoxyribonuclease 1 (APEX1) [181]. APEX1 is a multifunctional protein involved in DNA base excision repair and cellular response to oxidative stress, and it plays a key role in chondrocyte hypertrophy and osteoarthritis progression [181]. In preclinical models of osteoarthritis, intra-articular administration of siRNA targeting Apex1 has been shown to delay cartilage degradation, reduce proteoglycan loss, and decrease the expression of matrix metalloproteinase 13 (MMP13), suggesting its potential as a biological and pharmacological treatment for osteoarthritis [181].
