siRNA targeting ASNS is an experimental RNA interference (RNAi) therapeutic designed to silence the expression of asparagine synthetase (ASNS). ASNS is the enzyme responsible for the de novo synthesis of asparagine from aspartate and glutamine. In glioblastoma, tumor cells often exhibit a high demand for asparagine to support rapid proliferation, relying on both intracellular synthesis and extracellular uptake. Research conducted at Fujita Health University has demonstrated that genetic inhibition of ASNS using siRNA can suppress cell proliferation and induce apoptosis in both temozolomide-sensitive and temozolomide-resistant glioblastoma cell lines. This approach is being investigated as a potential metabolic therapy, particularly in combination with L-asparaginase, to exhaust the tumor's asparagine supply and overcome resistance to standard chemotherapies.
